Overview

Self-regulation is a multifaceted biological and psychological process rather than a single molecular entity, receptor, or enzyme. Biologically, the term refers to autoregulatory mechanisms, such as negative feedback loops in gene expression and autocrine signaling, which allow cells and organisms to maintain homeostasis and prevent excessive physiological responses [1, 5, 8]. For example, certain DNA- and RNA-binding proteins govern their own synthesis through direct feedback to ensure that cellular concentrations remain within a functional threshold [10, 14]. In a neuropsychological context, self-regulation encompasses the executive functions and emotional control mechanisms necessary for goal-directed behavior, primarily mediated by the prefrontal cortex and its associated neural circuits [2, 4, 13]. Although self-regulation is not a druggable molecule in the traditional sense, it represents a central therapeutic objective in psychiatry and behavioral medicine. Treatments for disorders characterized by self-regulatory dysfunction, such as ADHD or addiction, involve the pharmacological modulation of neurotransmitter systems (e.g., dopamine, norepinephrine, or serotonin) to stabilize and enhance these regulatory capacities [11, 13].

Mechanism of action

Pharmacological interventions do not target 'self-regulation' as a single molecule; instead, they modulate underlying neurotransmitter systems (e.g., through inhibition of dopamine and norepinephrine transporters) or nuclear receptors (e.g., PPARs) to restore or enhance endogenous regulatory feedback loops and cognitive control mechanisms [8, 11, 13].
