Overview

Empty spiracles homeobox 2 is a homeodomain transcription factor encoded by the EMX2 gene in humans. It is a critical regulator of brain patterning and neocortical area specification during embryonic development, and also participates in neural stem cell regulation, urogenital system formation, and inner ear development. EMX2 exerts its effects mainly through precise spatial and temporal gene regulatory networks, integrating extracellular signaling pathways (WNT, BMP, FGF8), repressing or activating target genes responsible for cell proliferation, migration, and differentiation. Disease implications are significant: loss or mutation of EMX2 is linked to structural brain malformations such as schizencephaly, developmental disorders, and cancer—especially via a tumor suppressor role with decreased expression driving proliferation and invasiveness. EMX2's function and modulation remain of high interest in neurodevelopmental biology and oncology.

Mechanism of action

Therapeutic restoration or enhancement of EMX2 function may suppress tumor proliferation and invasion, sensitize cells to cancer treatment, and inhibit oncogenic signaling pathways (e.g., canonical Wnt pathway). Silencing or loss of EMX2 generally increases proliferative and invasive phenotypes in cancer models.
