Overview

Endogenous retrovirus group H member 1 (ERVH-1 or HERV-H4p15.2) is a human genomic locus derived from ancient retroviral insertions. It belongs to the HERV-H family, characterized by long terminal repeats (LTRs) and sequences analogous to retroviral gag, pol, and env genes, though most protein-coding capacity has been lost. The locus primarily produces noncoding RNAs and functions as a transcriptional regulatory region. Its expression is epigenetically regulated and has been observed to be silenced in colon tumor tissues while being active in the corresponding normal tissues, suggesting a role in the regulatory landscape of cancer biology. While ERVH-1 is not targeted by current therapeutics and does not encode functional proteins, its regulatory activity through noncoding RNA and LTRs may impact host gene expression and epigenetic states
