Overview

Endogenous retrovirus group V member 1 envelope protein (ERVV-1) is a product of a human endogenous retrovirus (HERV) locus on chromosome 19. ERVV-1 is an envelope protein related to retroviral Env proteins, specifically integrated into the human genome long ago through infection of germline cells. In humans, ERVV-1 is highly expressed in the placenta, especially in syncytiotrophoblasts, and belongs to a broader group of envelope proteins collectively known as "syncytins" that in some mammals mediate cell–cell fusion critical for placental development[1][2][6]. However, in humans, ERVV-1 encodes a truncated envelope protein that has lost the canonical fusogenic (cell fusion-promoting) activity seen in some related genes (e.g., ERVW-1, ERVFRD-1)[1][2]. Although it no longer mediates cell fusion, its persistent expression suggests a possible, as-yet-undefined, regulatory or modulatory role in placental biology[1]. ERVV-1 does not appear to serve as a therapeutic target, nor is it currently implicated in specific disease processes. There are no known approved or investigational drugs that interact with ERVV-1, and it is not established as a disease biomarker.
