Overview

Endogenous tissue regeneration refers to the body’s intrinsic ability to repair and replace damaged tissues and organs using its own resident stem and progenitor cells, often coordinated and modulated by the local immune milieu and biochemical cues such as cytokines, growth factors, and changes in the tissue environment[1][2][3][4]. Unlike strategies that rely on transplantation of stem cells or engineered tissues, this approach leverages mechanisms like the activation of local stem cell populations (e.g., muscle satellite cells, mesenchymal stem cells), modulation of macrophage polarization, secretion of factors that promote proliferation and differentiation, and remodeling of the extracellular matrix[1][2][3][4]. While pivotal for health and recovery from injury, this process is not a defined molecular target but a complex, multi-factorial regenerative phenomenon, and is therefore not strictly considered a “therapeutic target” like a receptor or enzyme[1][2][3][4]. Most therapeutic interventions in this area have focused on biomaterials, biologics, or pharmacological agents that enhance or modulate endogenous regenerative pathways, rather than directly targeting a single molecule or protein[1][4].
