Overview

Endoglin (ENG) mRNA is the transcript encoding a 180 kDa homodimeric transmembrane glycoprotein that functions as a co-receptor for the transforming growth factor-beta (TGF-beta) receptor complex. It is predominantly expressed in vascular endothelial cells and is essential for maintaining vascular integrity and promoting angiogenesis (UniProt P17813). The mRNA itself serves as a therapeutic target for gene-silencing technologies, such as small interfering RNAs (siRNAs) and antisense oligonucleotides (ASOs), which aim to downregulate Endoglin protein expression in pathological states (PubMed: 30154014). In cancer, high levels of Endoglin are associated with increased tumor vascularization and poor prognosis, making the degradation of its mRNA a strategy to inhibit tumor growth and metastasis (PubMed: 28651611). Conversely, mutations in the ENG gene that lead to reduced mRNA stability or protein function cause Hereditary Hemorrhagic Telangiectasia type 1 (HHT1), a disorder characterized by arteriovenous malformations (PubMed: 25239236). Targeting Endoglin mRNA provides a precise method to modulate TGF-beta signaling, though challenges include ensuring endothelial-specific delivery and avoiding systemic vascular toxicity (PubMed: 21833013).
