Endoplasmic reticulum-Golgi intermediate compartment protein 1 (ERGIC1) is a cycling membrane protein localized at the ER-Golgi intermediate compartment, playing a critical role in the regulation of protein trafficking between the endoplasmic reticulum and Golgi apparatus[1][2][3][4][5]. It acts as a modulator in cycling protein complexes, stabilizing proteins such as hErv46, thus ensuring efficient cargo sorting and transport[1][2][4]. Dysregulation or mutation of ERGIC1 impairs developmental protein trafficking and is implicated in diseases such as congenital arthrogryposis multiplex congenita, certain cancers (including prostate and gastric), and potentially neurodegenerative conditions[1][3]. There is, to date, no established evidence that ERGIC1 is a therapeutic target for direct drug intervention, nor are there specific drugs, mechanisms of action, or biomarkers established for this protein[1][3][4].
