Overview

Endoplasmic reticulum junction formation protein lunapark (LNPK) is a conserved ER-shaping membrane protein characterized by two transmembrane domains and a zinc finger motif that is essential for its function. LNPK localizes specifically to three-way junctions of the ER network, where it stabilizes nascent junctions and modulates ER morphology by promoting network stability and limiting excessive junction mobility. Loss or depletion of LNPK disrupts ER morphology, shifting the network from a reticulated structure toward expanded sheets, and is associated with neurodevelopmental disorders such as epilepsy and hypoplasia of the corpus callosum. Although its direct role as a therapeutic target is not established, LNPK is essential for cellular homeostasis and proper ER function[1][2][3][5].
