Endoplasmic reticulum lectin 1 (ERLEC1) is a resident ER protein that binds to unfolded or misfolded glycoproteins through recognition of N-linked glycans, serving a critical function in ER-associated degradation (ERAD), which eliminates defective proteins from the secretory pathway. It interacts as part of multi-protein complexes to facilitate the transport and disposal of aberrant glycoproteins, thus maintaining cellular proteostasis; dysregulation of ERLEC1 or ERAD components is implicated in diseases featuring protein misfolding and aggregation, including neurodegenerative conditions and cancer. ERLEC1 is widely expressed, notably residing in the endoplasmic reticulum lumen, and although its role in disease is under investigation, it is regarded as a molecular cofactor and potential target in ER stress-associated pathologies.
