Overview

Endoplasmic reticulum membrane protein complex subunit 3 antisense RNA 1 (EMC3-AS1) is a long non-coding RNA (lncRNA) situated on chromosome 3q25 and produced in antisense orientation to the EMC3 gene. EMC3-AS1 is significantly upregulated in liver cancer tissues compared to normal liver tissues and is associated with poor prognosis. Its elevated expression correlates with increased proliferative and migratory capacity of liver cancer cells, as well as changes in immune infiltration and immune checkpoint gene expression. Silencing of EMC3-AS1 via siRNA in liver cancer cell lines inhibits cell proliferation and migration, suggesting potential as both a diagnostic/prognostic biomarker and a therapeutic target in liver cancer. It is implicated in gene regulatory processes and may modulate mRNA modifications such as N6-methyladenosine (m6A), but the exact molecular mechanisms in disease remain under investigation[1].

Mechanism of action

Targeting EMC3-AS1 by RNA interference (e.g., siRNA knockdown) suppresses proliferation and migration of liver cancer cells[1]. Theoretical mechanisms may involve modulating mRNA modifications (such as m6A), but specific drug mechanisms not established in current sources[1].
