Endosomal transmembrane epsin interactor 1 (ENTREP1) is a protein-coding gene involved in cellular trafficking and signaling regulation. It functions as an activator of the E3 ubiquitin ligase ITCH and is a key effector in the ubiquitination of the CXCL12-activated CXCR4 receptor. This process leads to endocytosis and desensitization of CXCR4, thereby regulating the CXCL12/CXCR4 signaling axis, which is important in cellular migration, immune response, and cancer progression. ENTREP1 is localized to the endosome and plasma membrane and participates in adaptation of signaling pathways. Diseases associated with mutations or dysregulation of ENTREP1 include prostate cancer and progressive leukoencephalopathy with ovarian failure. Its role as an endocytic adaptor protein relates to its capability to modulate cell surface receptor turnover and cell signaling.
