Endothelial activities encompass the diverse physiological and biochemical processes performed by the endothelial cell layer to maintain vascular health and systemic homeostasis. These activities include the regulation of vascular tone through the production of vasodilators like nitric oxide, the control of blood-tissue permeability, the modulation of immune cell adhesion, and the maintenance of a non-thrombogenic surface (Pober & Sessa, 2007; Nature Reviews Immunology). Dysfunction in these activities, often termed endothelial dysfunction, is a critical precursor to cardiovascular diseases, including atherosclerosis and hypertension, and plays a major role in diabetic microvascular complications (Deanfield et al., 2007; Circulation). While 'endothelial activities' is a functional descriptor rather than a specific molecular target, many therapeutic agents aim to improve these processes by acting on specific proteins such as eNOS, ACE, or VEGFR. Consequently, monitoring these activities is essential for assessing vascular health and the efficacy of treatments for inflammatory and ischemic conditions. Because this term refers to a category of biological processes rather than a single receptor or enzyme, it is considered an incorrect designation for a specific molecular drug target (StatPearls, 2023).
Drugs modulate endothelial activities by enhancing nitric oxide (NO) bioavailability, inhibiting vasoconstrictors like angiotensin II or endothelin-1, or blocking pro-angiogenic growth factors to restore vascular homeostasis and barrier integrity (Gimbrone & García-Cardeña, 2016; Circulation Research).
