Endothelial cell cytoskeleton

Molecular classification
Other (cellular structure), Cytoskeletal protein network (includes actin filaments, microtubules, intermediate filaments)
01

Overview

The endothelial cell cytoskeleton is a dynamic, interconnected system of protein filaments—including actin filaments, microtubules, and intermediate filaments—that supports the structure, motion, and specialized functions of vascular endothelial cells. It regulates cell shape, adhesion, migration, and is essential for maintaining the endothelial barrier, controlling vascular permeability and responding to physiological forces such as shear stress. Remodeling of the cytoskeleton underlies key processes in vascular inflammation, repair, angiogenesis, and is implicated in the pathogenesis of cardiovascular and inflammatory diseases.

02

Mechanism of action

Microtubule stabilization (prevents barrier-disruptive cytoskeletal remodeling; e.g., taxanes) Actin cytoskeleton modulation (altering actin polymerization and contractility affects endothelial barrier integrity and permeability)

03

Biological functions

Maintenance of cell shape and structural integrityRegulation of endothelial barrier function and permeabilitySignal transduction of mechanical and chemical stimuliCell migration, adhesion, and vascular remodeling
04

Disease associations

Cardiovascular disease (barrier dysfunction, atherosclerosis, thrombosis)Inflammation (endothelial hyperpermeability)Cancer (angiogenesis and metastasis)Other (edema, tissue injury, microvascular disease)
05

Safety considerations

Non-specificity: Drugs targeting the cytoskeleton often affect many cell types, causing systemic toxicitiesDisruption of vascular barrier may result in edema, bleeding, or organ dysfunction
06

Interacting drugs

Paclitaxel (stabilizes microtubules)
