Overview

Endothelial cell membrane components refer to the diverse set of proteins, glycoproteins, and lipids expressed on the surface of endothelial cells that line the entire vascular system. These components, which include receptors like Vascular Endothelial Growth Factor Receptor 2 (VEGFR2), adhesion molecules such as Intercellular Adhesion Molecule 1 (ICAM-1), and various integrins, are essential for maintaining vascular homeostasis and regulating blood pressure (PMID: 15159440). They also control the passage of fluids, solutes, and immune cells from the blood into the surrounding tissues. In many diseases, particularly cancer and chronic inflammation, these membrane components are dysregulated or overexpressed to facilitate pathological processes like tumor angiogenesis or excessive leukocyte infiltration (PMID: 21854926). Therapeutic strategies targeting these components aim to inhibit vessel growth in tumors or block inflammatory cell recruitment in autoimmune disorders. However, because the endothelium is ubiquitous and performs vital physiological functions, drugs targeting these components often carry risks of systemic side effects, including hypertension, hemorrhage, and impaired tissue repair (PMID: 25614462).
