Endothelial differentiation-related factor 1 (EDF1) is a ubiquitously expressed protein that acts primarily as a transcriptional coactivator. It stimulates the transcriptional activities of nuclear receptors such as NR5A1, LXRA (NR1H3), and PPARG, and enhances the DNA-binding activity of several transcription factors, including ATF1, ATF2, CREB1, and NR5A1[1][6]. EDF1 contains a cro/C1-type helix-turn-helix (HTH) DNA-binding domain and also binds calmodulin, thereby modulating nitric oxide synthase activity in the cytoplasm[2][6]. The protein is involved in processes such as endothelial cell differentiation, lipid metabolism, and hormone-induced cardiomyocyte hypertrophy[1][2][5][6]. Diseases associated with EDF1 include some forms of hereditary deafness[1][5]. EDF1 is not a therapeutic receptor, enzyme, transporter, or common drug target, and there are currently no known drugs that interact specifically with EDF1.
