Overview

Engager E1 is a novel, proprietary immune proximity engager developed by Dualitas Therapeutics for the treatment of autoimmune and allergic diseases. It serves as a standardized, reusable antibody arm that can be integrated into various bispecific antibody candidates to leverage cell-surface proximity biology. By binding to a specific immune cell receptor, E1 facilitates the co-localization of that receptor with a therapeutic target, thereby amplifying the drug's inhibitory or regulatory signals. This approach is designed to overcome the limitations of traditional monoclonal antibodies, offering potentially greater potency and faster onset of action. E1 is currently utilized in lead preclinical programs such as DTX-102 for rheumatoid arthritis and DTX-103 for asthma, where it aims to differentiate from existing validated therapies through its unique proximity-driven mechanism of action.

Mechanism of action

Engager E1 is a proprietary, reusable antibody arm that functions as a proximity-based trigger. It is designed to be 'plugged and played' into bispecific antibody (BsAb) formats, where it binds to a specific, undisclosed immune cell surface receptor to induce proximity between that receptor and a disease-relevant target (e.g., TNF or IgE). This proximity mechanism is intended to enhance pharmacologic activity, such as increasing potency or efficacy (Emax), beyond what can be achieved by conventional competitive inhibition. The E1 arm is engineered to be inert as a monovalent monospecific agent, only becoming active upon bivalent or bispecific engagement, which provides a safety window by preventing off-target activation.
