Overview

The Engineered Mechanosensitive Channel (EMC) is a synthetic or modified ion channel, typically derived from the bacterial Mechanosensitive Channel of Large Conductance (MscL), designed for therapeutic applications in mammalian cells. It functions as a biological "pressure-relief valve" or "nanovalve" that opens a large, non-selective pore in response to specific mechanical stimuli, such as increased membrane tension or acoustic pressure from ultrasound. In the context of glaucoma, EMC is delivered via gene therapy to the trabecular meshwork, where it senses elevated intraocular pressure and facilitates the drainage of aqueous humor to lower pressure autonomously. Beyond ophthalmology, EMC variants are a cornerstone of sonogenetics, allowing for the non-invasive modulation of specific neural circuits using low-intensity ultrasound. The channel's gating properties can be further tuned through site-directed mutagenesis to respond to other triggers like pH or light, making it a versatile tool for controlled drug delivery and synthetic biology. However, therapeutic use must address potential challenges such as the immunogenicity of the bacterial protein and the risk of spontaneous gating leading to cellular toxicity.
