Overview

Homeobox protein engrailed-1 (EN1) is a transcription factor that contains a homeodomain, enabling it to bind specific DNA sequences and regulate gene expression. EN1 is crucial for proper formation of the apical ectodermal ridge—a key structure in limb development— and for dorsal-ventral patterning in the limb. It controls pattern formation during development of the central nervous system and regulates transcription of specific genes, such as utrophin. EN1 belongs to the homeobox family of transcription factors, is highly conserved, and is implicated in several developmental diseases and cancers.
