Overview

Enhanced green fluorescent protein is an engineered variant of the original wild-type green fluorescent protein from the jellyfish Aequorea victoria. EGFP is notable for the F64L and S65T mutations, which improve its folding and fluorescence properties at 37°C compared to the wild-type protein. Like all GFPs, EGFP forms a beta-barrel fold (~239 amino acids, 26.9 kDa), enclosing an internal chromophore derived from cyclization and oxidation of Thr65, Tyr66, and Gly67[1][2][5][7]. Its main function in research is as a genetically encoded marker and reporter: it emits bright green fluorescence when exposed to blue or ultraviolet light, making it a ubiquitous tool for studying protein localization, gene expression, and cell lineage by fluorescence microscopy[2][5][6][7]. EGFP is not a human or animal protein target—such as a receptor, enzyme, transporter, or disease-related molecule—so it does not qualify as a therapeutic target. Its use is restricted to biotechnology, molecular biology, and imaging applications, with no established disease roles, interacting drugs, relevant mechanisms of action for drug targeting, biomarker applications, or associated safety/therapeutic concerns.
