Overview

Enhanced Green Fluorescent Protein (EGFP) is a synthetic derivative of the naturally occurring Green Fluorescent Protein (GFP) from the jellyfish Aequorea victoria [1]. It contains specific amino acid substitutions, notably F64L and S65T, which enhance its brightness, photostability, and folding efficiency at 37 degrees Celsius compared to the wild-type protein [1, 3]. EGFP is primarily used as a reporter gene product in molecular biology to monitor gene expression, protein localization, and cellular processes in real-time [2]. It is not a therapeutic target for drug development; instead, it serves as a vital research tool for visualizing the success of gene delivery systems and tracking the fate of engineered cells, such as CAR-T cells, in preclinical studies [2, 3]. While it is widely used in laboratory settings, its use in human clinical applications is limited by its potential to elicit an immune response as a non-human protein [2]. Consequently, EGFP is categorized as a diagnostic or experimental tool rather than a mediator of disease or a site for pharmacological intervention.
