Enhancer of mRNA decapping protein 3 (EDC3) is a critical scaffold protein that facilitates the assembly and activation of the mRNA decapping complex, primarily by interacting with the decapping enzyme DCP2 and its coactivator DCP1 (UniProt Q96F86). It plays a central role in the 5'-to-3' mRNA decay pathway, a major regulatory mechanism for controlling gene expression through the modulation of mRNA stability (NCBI Gene 80153). EDC3 is also a key component required for the formation of processing bodies (P-bodies), which are cytoplasmic granules where non-translating mRNAs are sequestered, stored, or degraded (PubMed 18305152). Clinically, homozygous mutations in the EDC3 gene have been linked to autosomal recessive intellectual disability and microcephaly, highlighting its essential function in human brain development (OMIM 609842). While there are currently no pharmacological agents targeting EDC3 in clinical use, its role in regulating the degradation of specific oncogenic or viral transcripts makes it a potential target for future therapeutic intervention in oncology and virology (PubMed 25533962).
