The enkephalin-dependent opioid pathway refers to the modulation of physiological and behavioral processes by enkephalins, which are endogenous opioid peptides. Enkephalins primarily act as neurotransmitters and neuromodulators in the central nervous system (CNS) and peripheral tissues, exerting their effects through binding to specific opioid receptors. This pathway is involved in pain modulation, emotional regulation, neuroprotection, and reward processing.
Enkephalins bind to opioid receptors (primarily Delta, Mu, and Kappa), leading to G-protein activation, inhibition of adenylyl cyclase, modulation of ion channels (increased K+ efflux, reduced Ca2+ influx), and ultimately, reduced neuronal excitability and neurotransmitter release.
