Overview

Enkurin domain containing 1 (ENKD1) is a microtubule-associated protein found in the cytoskeleton at centrosomes and cilia. It binds alpha-tubulin and plays pivotal roles in organizing microtubule-based structures, stabilizing microtubules, promoting proper mitotic spindle orientation during cell division, and enabling the assembly of non-motile (primary) cilia and the regulation of ciliary signaling. ENKD1 facilitates ciliogenesis by promoting the removal of CP110 from the mother centriole through competitive interaction, antagonizing CEP97. This is crucial for initiating cilium formation and impacts developmental processes such as left–right asymmetry. Dysregulation of ENKD1 has been associated with impaired asymmetric division in keratinocytes, defective epidermal layer stratification, and reduced ciliary signaling. Lower ENKD1 expression or gene fusion events have been implicated in cancer progression (notably non-small cell lung cancer and T-cell acute lymphoblastic leukemia), making ENKD1 a candidate biomarker and possible drug target.
