Enolase 4 (ENO4) is a member of the enolase family of enzymes that catalyze the conversion of 2-phospho-D-glycerate to phosphoenolpyruvate in the glycolytic pathway, an essential step for cellular energy production[2][3]. ENO4 is predicted to contribute to phosphopyruvate hydratase activity and is primarily implicated in glycolysis, as well as involved in the assembly and organization of cilia and the motility of flagellated sperm, with localization to the sperm principal piece in humans[2][3]. ENO4 is considered important for energy metabolism and may play roles in specialized biological processes, such as tissue or organ regeneration in certain invertebrate models (e.g., siphonal regeneration in bivalve mollusks), but in humans, its principal characterized function is linked to sperm function[1][2][3]. Mutations in ENO4 have been associated with Immunodeficiency 24, a rare human disease[2]. There are currently no approved drugs known to target ENO4 specifically, nor are there well-established clinical biomarkers or safety concerns directly linked to selective modulation of ENO4 function[2].
