Enoyl-CoA delta isomerase 2 (ECI2) is an enzyme localized primarily to the mitochondria and peroxisomes, belonging to the hydratase/isomerase superfamily[1][2]. It is a critical catalyst in the beta-oxidation of unsaturated fatty acids, specifically mediating the isomerization of 3-cis and 3-trans enoyl-CoA esters to 2-trans-enoyl-CoA intermediates, a necessary step in the mitochondrial and peroxisomal degradation of cis-, mono-, and polyunsaturated fatty acids[1][2][3]. ECI2 enables tissues such as the heart, liver, and steroidogenic organs to efficiently metabolize fatty acids for energy production, lipid homeostasis, and steroid biosynthesis. Alternative splicing results in multiple isoforms with tissue-specific functions, including the formation of peroxisome–mitochondria contact sites supporting steroid production in Leydig cells[1]. ECI2 expression is linked to disease; it is upregulated in several cancers, possibly contributing to altered lipid metabolism characteristic of proliferating tumor cells, and is implicated in inherited peroxisomal metabolic disorders[1][2].
