The Enterovirus A71 virion is a non-enveloped, icosahedral viral particle about 20–30 nm in diameter, composed of 60 protomers, each containing four structural capsid proteins (VP1, VP2, VP3, VP4). The capsid encloses a single-stranded positive-sense RNA genome (~7,500 nt). Surface proteins VP1, VP2, and VP3 mediate host cell attachment primarily through the VP1 "canyon" region, interacting with host receptors such as SCARB2 and PSGL-1. Various neutralizing epitopes are exposed on VP1 and VP2, making the virion both a direct therapeutic and vaccine target. Neutralizing monoclonal antibodies and vaccines (especially those targeting VP1) have been developed to inhibit viral entry and propagation, with ongoing challenges related to genetic diversification and neurovirulence[1][2][3][4][5][6][7][8].
