Overview

The Enterovirus A71 (EV-A71) capsid protein VP1 GH loop is a critical surface-exposed structural element that plays a central role in viral pathogenesis and host cell entry (PubMed: 23552837). It is primarily responsible for mediating the attachment of the virus to its cellular receptors, most notably Scavenger Receptor Class B Member 2 (SCARB2) and P-selectin Glycoprotein Ligand-1 (PSGL-1), which are essential for viral internalization and uncoating (PubMed: 19575010, 19543277). Due to its high exposure on the viral surface, the GH loop is a major neutralizing epitope and a primary target for the development of vaccines and therapeutic antibodies (PubMed: 25855244). Inactivated EV71 vaccines, which are used to prevent hand, foot, and mouth disease (HFMD), rely heavily on the induction of antibodies targeting this region (PubMed: 24580462). Mutations within the GH loop can lead to significant changes in viral virulence, tissue tropism, and the ability to escape host immune responses, making it a key focus for molecular surveillance (PubMed: 22114482). Therapeutic strategies targeting this loop aim to block receptor binding sites, thereby neutralizing the virus before it can infect host cells.
