The protein is characterized by the presence of an Epsin N-terminal homology (ENTH) domain, a module implicated in endocytosis and membrane trafficking[4][2][11]. ENTHD1 is expressed across multiple human organs, with highest levels in the testes[1][3]. Functional data are limited, but bioinformatic predictions and analogy to other ENTH domain proteins (such as Epsin) suggest a role in clathrin-mediated endocytosis and possibly in actin cytoskeleton dynamics[1][4][6][11]. There are no known therapeutic agents targeting this protein, and it does not currently feature as a validated disease target or biomarker. Some aliases, particularly CACNA1I, are likely erroneous or due to earlier annotation errors[1][5].\n\nNote on aliases: ENTHD1 is sometimes inaccurately listed as "CACNA1I," which strictly refers to a gene encoding a T-type voltage-gated calcium channel subunit. In this context, ENTHD1 should not be confused with CACNA1I[1][3][5].
