Overview

ENTPD3 antisense RNA 1 (commonly abbreviated as ENTPD3-AS1, also known as EIF1B-AS1) is a long non-coding RNA (lncRNA) transcribed antisense to the ENTPD3 locus and previously referred to as EIF1B antisense RNA 1 in some annotations[1][2][3]. This molecule does not encode a protein and functions at the RNA level. Several studies indicate that ENTPD3-AS1 acts as a regulatory RNA in cancer. For example, in lung adenocarcinoma, ENTPD3-AS1 is downregulated in tumor tissue, with low expression associated with poor prognosis. Functional studies suggest ENTPD3-AS1 acts as a tumor suppressor lncRNA, in part by acting as a “sponge” for microRNAs such as miR-421, thereby de-repressing downstream tumor suppressors like NR3C1 and potentially regulating processes such as migration and epithelial-mesenchymal transition (EMT)[3]. In breast cancer, including metastatic triple-negative cases, ENTPD3-AS1 (referred to as EIF1B-AS1 in some studies) is also downregulated in tumors compared to normal or non-metastatic tissue, suggesting a role in metastasis and disease progression, and current data suggest this lncRNA could serve as a diagnostic and prognostic biomarker in these settings[2]. The exact molecular mechanism of ENTPD3-AS1 remains under active investigation, and no approved drugs are currently known to target this RNA directly. Its role is primarily as a biomarker rather than a direct therapeutic target.
