Overview

Envelope glycoprotein G-2 (gG-2) is a membrane-associated protein specific to Herpes Simplex Virus type 2 (HSV-2), encoded by the US4 gene (UniProt: P06484). It serves as a critical tool in clinical diagnostics, as it allows for the serological differentiation between HSV-1 and HSV-2 infections due to its unique antigenic properties (CDC: STI Guidelines). Biologically, gG-2 acts as a viral chemokine-binding protein (vCKBP), which binds to various human chemokines such as CXCL12 and CCL5 (PubMed: 16148008). This binding activity disrupts the normal recruitment of leukocytes to the site of infection, thereby facilitating viral immune evasion and persistence. In the context of drug development, gG-2 has been utilized as a key antigen in subunit vaccines like GEN-003, designed to stimulate both humoral and cell-mediated immunity (PubMed: 28137893). These therapeutic approaches aim to reduce the frequency of viral shedding and the recurrence of clinical lesions in infected individuals. However, the development of effective gG-2-targeted therapies is hindered by the virus's ability to establish permanent latency in the host's nervous system. Furthermore, the complex interactions between gG-2 and the host immune system present significant challenges for achieving complete sterilizing immunity.
