The envelope glycoprotein gp41 is a critical component of the HIV-1 envelope spike complex, which also includes gp120. Together, these proteins mediate the virus's entry into host cells. Gp41 is the transmembrane subunit responsible for catalyzing fusion between the viral and cellular membranes after initial attachment by gp120. After HIV binds to CD4 and coreceptor on target cells via gp120, conformational changes expose and activate gp41. Gp41 undergoes dramatic structural rearrangements to facilitate membrane fusion.
