The V3 loop is the third variable region (V3) of the HIV-1 envelope glycoprotein gp120, a critical component of the viral spike that mediates entry into host cells. The V3 loop is located on the outer domain of gp120 and typically consists of 33–35 amino acids, bounded by two invariant cysteines that form a disulfide bond to create its characteristic loop structure. It plays a crucial role in coreceptor binding (CCR5 or CXCR4), determines viral tropism, and is a major target for neutralizing antibodies. However, its high sequence variability and conformational flexibility pose significant challenges for vaccine development.
