Overview

ENY2 is a small, evolutionarily conserved protein integral to nuclear gene expression regulation. It serves as a core adaptor subunit in the SAGA/TFTC deubiquitination module, essential for histone H2A and H2B deubiquitination, which counteracts heterochromatin silencing and facilitates nuclear receptor‐dependent transcription. Additionally, ENY2 forms part of the TREX-2 complex regulating mRNA export through nuclear pores. It is recruited to transcribed genes and interacts with various transcription-regulatory, insulator, and export proteins. ENY2's loss or dysfunction can lead to impaired transcription, defective mRNA export, and developmental abnormalities, implicating it in diverse cellular processes and disease states
