Overview

EP300-interacting inhibitor of differentiation 3 (EID3) is a multifunctional nuclear protein that acts as a transcriptional corepressor, primarily through interactions with EP300/CREBBP, and as a component of the SMC5-SMC6 complex involved in homologous recombination-mediated DNA double-strand break repair[1][2][5][7]. EID3 modulates chromatin structure and epigenetic modifications, regulates differentiation (notably in neural and germ cell lineages), and is required for proper meiosis and chromatin condensation during spermatogenesis[1][3][4]. Overexpression is linked to cancers such as glioblastoma, where it predicts poor prognosis, and it may act as a therapeutic target by sensitizing resistant cancer cells to radiation and chemotherapy[1]. EID3 is also associated with rare diseases including Sotos syndrome 2 and muscular dystrophy, and is being studied as a potential biomarker and immunological target in multiple cancer types[2][4].
