Overview

Ephedradines A-D are a group of macrocyclic spermine alkaloids primarily found in the roots of Ephedra species, known as Ephedra Radix or "Mao-kon" in Traditional Chinese Medicine [1, 2]. Unlike the ephedrine alkaloids found in Ephedra stems, which stimulate the sympathetic nervous system and raise blood pressure, Ephedradines A-D are characterized by their potent hypotensive (blood-pressure-lowering) activity [5, 10]. Among these, Ephedradine B is the most active and lowers blood pressure by acting as a ganglion-blocking agent, inhibiting neurotransmission through autonomic ganglia [7, 10, 12]. These alkaloids also demonstrate anti-inflammatory and antioxidant properties, often by suppressing inflammatory signaling pathways such as NF-kappaB [10, 11]. Historically used to treat hypertension and excessive sweating, Ephedradines serve as natural lead compounds for the discovery of cardiovascular and anti-inflammatory therapeutics [4, 12, 14]. They represent a unique class of polyamine derivatives with multi-target pharmacological potential in traditional and modern medicine [9, 13].

Mechanism of action

Ephedradines A-D exert hypotensive effects primarily through the blockade of autonomic ganglia (antagonism of ganglionic nicotinic acetylcholine receptors) and the suppression of inflammatory mediators via inhibition of NF-kappaB activation [7, 10, 12].
