Overview

Ephrin-B3 is a cell surface transmembrane ligand for Eph receptors, the largest subfamily of receptor tyrosine kinases. Ephrin-B3 plays crucial roles in bidirectional cell signaling, especially during neural, vascular, and epithelial development. The protein is encoded by the EFNB3 gene and contains a transmembrane domain with a cytoplasmic tail that enables reverse signaling through phosphorylation and PDZ protein interactions. Ephrin-B3 participates in axon guidance, neurogenesis, synaptic plasticity, and has pivotal functions in the forebrain and other nervous system regions. Additionally, Ephrin-B3 can act as a receptor for Nipah and Hendra viruses. Dysfunction or alteration of EFNB3 signaling is implicated in diseases including craniofrontonasal syndrome, encephalitis, and potentially cancer or neurodevelopmental disorders.
