Overview

Ephrin type-A receptor 6 (EPHA6) is a member of the Ephrin receptor subfamily of receptor tyrosine kinases, encoded by the *EPHA6* gene[1][5][6][11]. EphA6 is a transmembrane protein that binds GPI-anchored ephrin-A ligands on adjacent cells, mediating contact-dependent, bidirectional signaling and playing critical roles in axon guidance and neural development[1][3][8][11]. Eph receptors, including EPHA6, are divided into subclasses A and B. EPHA6 is predominantly expressed in the brain and implicated in shaping neuronal morphology and spine development[7][8]. Although less characterized than other Eph receptors, EPHA6 has emerging roles in cancer biology, particularly breast carcinoma, where it may serve as a prognostic biomarker or therapeutic target, especially in hormone receptor-negative and HER2-positive subtypes[5]. The Eph family generally is targeted for drug development through kinase inhibition, but no specific drugs for EPHA6 have been approved; experimental evidence suggests that pseudokinase domains, like those of closely related EphA receptors, may bind ATP and be druggable[2].
