The skin barrier is a complex structural and functional unit of the skin, comprising primarily the outermost layer of the epidermis, the stratum corneum. It protects internal tissues from environmental threats (e.g., pathogens, toxins, UV light), prevents excessive loss of water (transepidermal water loss), and contributes to immune defense, microbial homeostasis, and maintenance of a healthy pH. It consists of corneocytes bound together with a lipid matrix of ceramides, fatty acids, and cholesterol, as well as other biochemical and immune components. Disruption leads to dryness, inflammation, increased risk of infection, and aggravated skin diseases, while many topical and systemic therapies aim to support or restore its integrity for both cosmetic and therapeutic benefit.
