Overview

Epidermal barrier repair is the process by which the skin restores its outer protective barrier—primarily the stratum corneum—to prevent water loss, block invading pathogens and chemicals, and maintain healthy skin homeostasis. The process is carried out by coordinated interactions among keratinocytes, immune cells, structural proteins (filaggrin, loricrin, involucrin), lipids (ceramides, cholesterol, free fatty acids), cell–cell junctions (tight junctions, desmosomes), and multiple signaling pathways. Disruption of this process underlies several inflammatory and infectious skin disorders. Therapeutic interventions are generally aimed at supporting or accelerating physiological repair rather than targeting a single defined molecule or receptor[1][2][3][4].
