EGFR806 refers to a unique conformational epitope on the extracellular domain of the epidermal growth factor receptor (EGFR) that is specifically recognized by the mAb806 antibody. This epitope is usually buried or not accessible in the normal, "tethered" form of EGFR on healthy tissues but becomes exposed when EGFR is overexpressed, misfolded, aberrantly glycosylated, or mutated—as seen in many cancers, particularly in tumors expressing the EGFRvIII mutant or amplified wild-type EGFR. Therapeutic approaches such as EGFR806-directed CAR T cells or antibody-drug conjugates target this epitope to achieve selective destruction of tumor cells while sparing most normal cells, reducing off-target toxicity compared with conventional anti-EGFR therapies.
