Epidermal growth factor receptor kinase substrate 8-like protein 3 (EPS8L3) is a member of the EPS8 family of signaling adaptor proteins that share similar structural domains, including an N-terminal phosphotyrosine-binding domain and an SH3 domain[2]. EPS8L3 is encoded by the *EPS8L3* gene in humans. It is functionally related to EPS8, a known substrate of the epidermal growth factor receptor (EGFR), but its precise molecular functions remain incompletely characterized[3]. Current evidence highlights that EPS8L3 is upregulated in liver cancer tissues and cell lines, where its overexpression promotes cancer cell proliferation and migration, likely via regulation of the PI3K/AKT signaling pathway[1]. High EPS8L3 expression is associated with higher tumor grade and poorer overall survival in liver cancer patients, and knockdown of EPS8L3 suppresses malignant cell growth and migration in vitro, suggesting it has oncogenic properties and may serve as an independent prognostic marker in liver cancer[1]. No drugs are currently known to directly target EPS8L3, and there is no established mechanism of action for drug interaction. EPS8L3 is best classified as a signaling adaptor rather than a classical therapeutic target such as a receptor, enzyme, or transporter; however, as research emerges, its role as a potential target in oncology may become clearer[1][2][5].
