Overview

Epidermal growth factor receptor messenger RNA (EGFR mRNA) is the intermediate nucleic acid molecule that carries genetic information from the EGFR gene to the ribosome for translation into the EGFR protein. As a member of the ErbB family of receptor tyrosine kinases, the resulting protein plays a pivotal role in signaling pathways that govern cell division, survival, and differentiation. In many oncogenic contexts, such as non-small cell lung cancer and glioblastoma, the EGFR gene is amplified or overexpressed, leading to an abundance of EGFR mRNA and a subsequent pathological increase in receptor density on the cell surface. Targeting the mRNA directly via antisense oligonucleotides (ASOs) or small interfering RNAs (siRNAs) offers a therapeutic strategy to silence the gene's expression at the pre-translational level. This approach is particularly valuable for potentially bypassing resistance mutations that render traditional small-molecule tyrosine kinase inhibitors (TKIs) ineffective, although clinical success depends heavily on overcoming delivery barriers to the tumor microenvironment.
