EPS15L1 is a modular cytosolic protein with multiple interaction domains, serving as an endocytic accessory mainly in clathrin-mediated and clathrin-independent endocytosis. It functions by linking cargo proteins and other adaptors to the endocytic machinery, helping internalize and sort receptors such as integrin beta-1 and transferrin receptor, which are important for iron homeostasis and cell adhesion. Genetic studies suggest it plays crucial, nonredundant roles in the nervous system and limb development, and its loss causes split-hand/split-foot malformations. EPS15L1 is related to, but distinct in function from, its paralog EPS15. Unlike membrane-spanning receptors or enzymes, EPS15L1 is primarily intracellular, forming protein complexes at membrane pits without direct ligand interaction, and is not currently a therapeutic target.
