Overview

The Epidermal growth factor receptor pathway substrate 8 (EPS8) peptide–HLA class I complex is a tumor-associated antigen complex that serves as a critical target for cancer immunotherapy. EPS8 is a signaling protein involved in actin remodeling and cell proliferation, which is frequently overexpressed in various malignancies, including acute myeloid leukemia and several solid tumors (PubMed: 12438259). In the context of the immune system, EPS8-derived peptides are processed and presented by HLA class I molecules on the surface of cells, including dendritic cells, where they are recognized by the T-cell receptors (TCRs) of CD8+ cytotoxic T lymphocytes (PubMed: 18436244). This recognition is a fundamental step in the activation of an adaptive immune response against cancer cells. Therapeutic approaches targeting this complex include the use of dendritic cell vaccines pulsed with EPS8 peptides and the development of TCR-engineered T-cell therapies (PubMed: 21149645). These strategies aim to exploit the differential expression of EPS8 between cancerous and normal tissues to achieve selective tumor destruction.
