Overview

Epidermal growth factor receptor truncated (EGFRt) refers to a modified form of EGFR where portions of the extracellular or intracellular domain have been removed or altered. While naturally occurring truncations in cancer can drive oncogenic signaling, engineered truncated forms are commonly used as non-signaling surface markers for tracking genetically modified cells, such as CAR-T cells. These engineered versions retain antibody recognition sites but lack functional kinase activity.

Mechanism of action

Varies depending on the specific truncation. Wild-type EGFR activates downstream signaling cascades including MAPK/ERK and PI3K/AKT pathways. Truncated forms, particularly engineered ones, are often designed to be non-signaling and used as surface markers. Naturally occurring truncations can exhibit constitutive activation.
