Target intelligence / Profile preview
Epidermal keratinocyte membrane lipids, primarily located within the stratum corneum, constitute the essential extracellular matrix that forms the skin's permeability barrier [StatPearls, 2023]. These lipids are organized into lamellar bilayers consisting of a specific ratio of ceramides, cholesterol, and free fatty acids [Journal of Investigative Dermatology, 2012]. Their primary biological function is to prevent excessive transepidermal water loss (TEWL) and protect the body from environmental insults, including pathogens and allergens [PubMed, 2020]. In various dermatological conditions such as atopic dermatitis and psoriasis, the composition and organization of these lipids are significantly altered, leading to barrier dysfunction [NIH, 2021]. Therapeutic interventions often involve the topical application of physiological lipid mixtures or occlusive agents to restore barrier integrity and alleviate symptoms of dryness and inflammation [Dermatologic Therapy, 2021]. Understanding the complex interplay of these lipids is crucial for developing effective treatments for skin barrier-related diseases.
Restoration of the skin's permeability barrier by replenishing essential lipids (ceramides, cholesterol, and fatty acids) or providing an occlusive layer to prevent transepidermal water loss (TEWL).
