Epididymal sperm-binding protein 1 (ELSPBP1) is a secreted sperm-coating glycoprotein expressed by the epididymal epithelium. Upon ejaculation, it binds to the surface of spermatozoa and is implicated in the process of sperm capacitation—the physiological changes sperm undergo for fertilization. ELSPBP1 contains four fibronectin type 2 (Fn2) domains in tandem, unique among such proteins. It exhibits heparin and phosphorylcholine binding properties, and has been shown to interact with biliverdin reductase A (BLVRA), possibly indicating additional roles in oxidative protection during sperm maturation, especially in bovine models. The protein is not classified as a receptor, enzyme, GPCR, transporter, or transcription factor, and is not considered a direct therapeutic target; its main biological relevance is in male reproductive physiology and sperm function. There are no known drugs targeting ELSPBP1 in clinical practice, nor established use in patient selection or disease monitoring.
