Overview

Epithelial barrier function refers to the collective physiological role played by epithelial tissues in forming a protective interface between the body and its environment. This includes the skin, gastrointestinal tract, respiratory tract, and other mucosal surfaces. The epithelial barrier is composed of closely packed cells connected by intercellular junctions such as tight junctions, adherens junctions, and desmosomes. These structures regulate permeability—allowing selective passage of nutrients or gases while preventing entry of pathogens or harmful substances—and maintain tissue integrity[1][3][5]. The epithelium also contributes to immune defense by secreting antimicrobial peptides and signaling molecules that modulate immune responses[2][6]. Disruption or dysfunction in this barrier is implicated in various diseases including infections, inflammatory conditions like atopic dermatitis or inflammatory bowel disease, and allergic disorders[1][6].
