The maintenance of this barrier involves a dynamic regulation of protein-protein interactions, such as those between claudins and occludins, which are often modulated by intracellular signaling pathways like the myosin light chain kinase (MLCK) pathway. In addition to physical sealing, the barrier function includes the secretion of mucus and antimicrobial peptides that form a biochemical shield.
