Overview

EMP3 is a 163-amino acid integral membrane protein (~18 kDa) encoded on chromosome 19q13.3. It structurally comprises four transmembrane domains, two extracellular loops (18 and 41 amino acids, respectively), and a short intracellular loop. EMP3 is classified as a tetraspanin membrane protein and shares homology with the PMP22 family, associated with myelin and membrane organization. Functionally, EMP3 regulates cell–cell interactions, signal transduction, and membrane organization, thereby influencing processes such as cell survival, migration, and tumor progression. It interacts with critical oncogenic membrane proteins (receptor tyrosine kinases, integrins, transforming growth factor beta receptor 2) to modulate cancer-relevant pathways (e.g., HER2/PI3K/Akt, TGF-beta/Smad, FAK/SRC, MAPK/ERK). EMP3's overexpression in tumors, particularly glioblastoma, is linked to poor prognosis and aggressive tumor phenotypes. It is also implicated as a regulator in immune cell biology
